Abstract
The transport of substrates across biological membranes can be broadly classified into translocation of unfolded proteins (both co-translational and post-translational), translocation of folded proteins (exclusively post-translational), and transport of small molecules. In the research work carried out as part of this thesis, structural and functional aspects of a translocase for unfolded proteins in Escherichia coli (SecYEG-YidC), a translocase of a folded protein in the Saccharomyces cerevisiae mitochondrial inner membrane (Bcs1) and a transporter of polysaccharides in Pseudomonas aeruginosa (PelBC) have been investigated, revealing novel aspects of their mechanism and offering new perspectives into their function in the context of the membranes where they reside.
