Abstract
This dissertation evaluates the therapeutic potential of newly synthesized histone deacetylase inhibitors (HDACi) for the treatment of diffuse large B-cell lymphoma (DLBCL). The study focused on the preclinical assessment of 41 HDACi across various lymphoma cell lines. Among these, YAK477 emerged as a particularly promising candidate, showing selective cytotoxicity in cell lines harboring CREBBP mutations. In addition to in vitro cytotoxicity, the tumor microenvironment was studied using co-culture systems with stromal cells.
Abstract
In dieser Dissertation wurde das therapeutische Potenzial neu entwickelter Histondeacetylase-Inhibitoren (HDACi) für die Behandlung des diffusen großzelligen B-Zell-Lymphoms (DLBCL) untersucht. Im Fokus stand die präklinische Analyse von 41 HDACi auf verschiedenen Lymphomzelllinien. Durch umfangreiche Viabilitätsassays und genetische Analysen konnte YAK477 als besonders vielversprechender Wirkstoff identifiziert werden, insbesondere in Zelllinien mit CREBBP-Mutationen. Neben zellulären Effekten wurde auch die Rolle der Tumormikroumgebung beleuchtet, wobei Ko-Kultur-Modelle mit Stromazellen zum Einsatz kamen.
